The housefly (Musca domestica) is an important host for a variety of bacteria, including some pathogenic and antibiotic-resistant strains. To further investigate the relationship between the housefly and the bacteria it harbors, it is necessary to understand the fate of microorganisms during the larval metamorphosis. The major bacterial communities in three developmental stages of the housefly (maggot, pupa, and adult fly) were investigated by a culture-independent method, polymerase chain reaction–denaturing gradient gel electrophoresis (PCR?DGGE) analysis of 16S rRNA genes. The bacteria that were identified using DGGE analysis spanned phyla Proteobacteria, Firmicutes, and Bacteroidetes. Changes in the predominant genera were observed during the housefly development. Bacteroides, Koukoulia, and Schineria were detected in maggots, Neisseria in pupae, and Macrococcus, Lactococcus, and Kurthia in adult flies. Antibiotic-resistant bacteria were screened using a selective medium and tested for antibiotic susceptibility. Most resistant isolates from maggots and pupae were classified as Proteus spp., while those from adult flies were much more diverse and spanned 12 genera. Among 20 tested strains across the three stages, 18 were resistant to at least two antibiotics. The free amino acids of potatoes irradiated with the doses of 7,000, 15,000 and 30,000 rad were determined by ion-exchange chromatography.After 15 days storage following irradiation, it was shown that the concentration of asparatic acid, proline and aliphatic amino acids increased with increasing irradiation doses, while that of basic amino acids and glutamic acid especially decreased. However, after 105 days of storage, the similarity of the free amino acid content of irradiated potatoes to that of non-irradiated and non-stored potatoes was observed.On the concentration of protein-bound amino acids, there were no significant differences between non-irradiated and 15,000 rad irradiated potatoes. 相似文献

A new antibiotic, toromycin (antibiotic B-21085) was isolated as yellow crystals from the culture broth of Streptomyces collinus subsp. albescens subsp. nov. The antibiotic has the molecular formula C27H26O9 and is the polycyclic aromatic hydrocarbon group of antibiotics. Changes in the nonprotein nitrogenous compounds produced from rabbit skeletal muscle (L. dorsi) by proteolysis were investigated.The value of trichloroacetic acid soluble nitrogen, ninhydrin positive materials and phenol reagent positive materials increased during storage at low and high temperature. Four components of ovomucoid were digested exhaustively and four kinds of glycopeptide corresponding to the four components were separated by gel filtration. Each glycopeptide was shown to be homogenious by paper chromatography and paper electrophoresis. Molar ratios of carbohydrate components of these glycopeptides varied to some extent but the amino acid compositions of these glycopeptides were essentially identical with each other with the exception of alanine. Aspartic acid and threonine were predominant amino acids in the all glycopeptides. It is most likely that the modes of linkages between polysaccharide and protein in individual ovomucoid I, II, III and IV are essentially the same, and that the carbohydrate moiety is linked to the protein via asparaginyl residue or the hydroxyl group of threonine, although the possibility of the linkages to glutamine and serine can not be excluded. 相似文献

Ethanol (1:1) extract of defatted soybean flour was fractionated systematically and the resulting phonolic acid fraction was investigated. This fraction had strong phenol-like flavor and contained at least seven phenolic acids including syringic, vanillic, ferulic, gentisic, salicylic, p-coumaric, and p-hydroxybenzoic acids. The main component among these was syringic acid, which was isolated as 3,5-dinitrobenzoate.In addition, two isomers of chlorogenic acids, presumably isochlorogenic and chlorogenic acids approximately in a ratio of 1 : 10, were found in this extract. Decarboxylation of about twenty kinds of α, β and γ-amino acids in the reaction with glyoxal or ninhydrin was investigated. The decarboxylation rate of amino acids proved that steric and polar effects had important roles in the reaction.From the data of pK2 values and decarboxylation rates of amino acids, it can be concluded that under a similar steric environment, the decarboxylation rate depends on the anion concentration of amino acids.Besides carbon dioxide, acetaldehyde, 2-propanone and propionaldehyde were respectively detected from the reaction of β-alanine, β and γ-amino-n-butyric acids with glyoxal or ninhydrin. From defatted n-paraffin-assimilating yeast cells, a crude protein was obtained by alkaliextraction followed by acid-precipitation. Then the protein was treated with ether until extractable substances were removed exhaustively at this stage. However, at the next stage where the ether-treated protein had been partially hydrolyzed with pepsin, when the hydrolysate was retreated with ether, it was found that ether-extractable substances totalling 270 mg/100 g were obtainable additionally. Chromatographic investigations demonstrated that the substances included significant amounts of aliphatic and aromatic hydrocarbons, some indoles, and a ubiquinone (n = 8).From the protein hydrolysate (substrate) after the above ether-treatment, a plastein was synthesized with Bioprase under the specific conditions. The plastein was obtained as a precipitate when the whole reaction mixture was treated with aqueous ethanol or acetone. The quantity and quality (nitrogen content) of the plastein depended on the ethanol or acetone concentration. Roughly speaking, the higher the concentration, the more the plastein quantity. The converse relation held for the quality; a plastein precipitated by treatment solely with water showed a higher quality than any other case. 相似文献
